Semax vs Selank: How the Research Compares
Semax and Selank are synthetic heptapeptides developed by Russian researchers. They share the same Pro-Gly-Pro tail but start from different parent peptides: Semax from the ACTH(4-7) fragment of adrenocorticotropic hormone, Selank from the immune peptide tuftsin. Semax research centers on BDNF and rodent ischemia models; Selank research centers on GABA receptor binding, enkephalin-degrading enzymes and stress behavior in rodents. Neither is approved by the FDA, and most of the evidence comes from a small group of Moscow institutions.
What are Semax and Selank?
Semax and Selank came out of the same Russian research network, centered on the Institute of Molecular Genetics of the Russian Academy of Sciences in Moscow. Both are heptapeptides, both end in Pro-Gly-Pro, and both were designed as hybrids: a short, biologically active fragment of a natural peptide joined to a glyproline tail1,2,3.
Glyprolines are proline- and glycine-containing peptides that Moscow physiologists reported to be far more stable in the body than typical regulatory peptides; adding Pro-Gly-Pro was meant to protect the active fragment from rapid breakdown3. Both peptides are still metabolized. Tritium-labeling studies identified His-Phe-Pro-Gly-Pro and Pro-Gly-Pro as the main products of Semax breakdown in the presence of nerve cells, and Thr-Lys-Pro-Arg-Pro, Thr-Lys-Pro, Arg-Pro and Gly-Pro as the main products of Selank breakdown in blood plasma4.
The difference lies in the front half. Semax begins with Met-Glu-His-Phe, the 4-7 segment of adrenocorticotropic hormone (ACTH), and is described as an analog of ACTH(4-10)1. Selank begins with Thr-Lys-Pro-Arg, the sequence of tuftsin, and is described as an analog of that immunomodulatory peptide5,6.

Semax research: BDNF and ischemia models
The core Semax finding is rapid induction of brain-derived neurotrophic factor (BDNF), a growth factor involved in synaptic plasticity. In the rat hippocampus, Semax raised BDNF protein, phosphorylation of the BDNF receptor TrkB, and BDNF and TrkB mRNA, and treated rats made more conditioned avoidance responses in a learning task1.
In rat models of stroke-like injury, Semax increased transcription of neurotrophin genes and their receptors in ischemic cortex after permanent middle cerebral artery occlusion, whereas the Pro-Gly-Pro fragment alone acted mainly non-specifically7. After transient occlusion with reperfusion, RNA sequencing found that Semax suppressed inflammation-related genes and activated neurotransmission-related genes, roughly reversing the pattern caused by the injury itself8. These studies measure gene expression at set time points in rats; they do not establish long-term functional outcomes.
Selank research: GABA, enkephalins and stress models
Selank's mechanism has been approached from several angles. Radioligand studies on brain cell membranes suggest it acts as a positive allosteric modulator of GABA receptors and that it alters the binding effects of diazepam and olanzapine2. Earlier work showed that Selank inhibited enkephalin-degrading enzymes in human plasma more potently than bacitracin or puromycin; the researchers proposed this as a possible basis for the anxiolytic-like activity reported in animal studies9.
Selank has also been studied in rodent stress and exposure models. In rats given ethanol long term, it prevented ethanol-related changes in hippocampal and prefrontal BDNF content and the memory deficits seen in an object recognition test during withdrawal6. So BDNF appears in both literatures, but in different models: healthy rat brain for Semax, an alcohol-exposure model for Selank.
Where Semax and Selank overlap
The two peptides have been tested side by side in a few studies. In the same in vitro experiment, both inhibited enkephalin-degrading enzymes in human serum, with Semax slightly more potent; pentapeptide fragments of each retained activity, while tri-, tetra- and hexapeptide fragments did not10.
In a rat model of Parkinson-like dopamine-neuron damage induced with 6-OHDA, neither peptide changed motor activity or passive defensive behavior, but Selank reduced anxiety-like behavior in an elevated maze, matching earlier findings in healthy rodents5. In people, a resting-state fMRI study in 52 healthy participants scanned the brain before and 5 and 20 minutes after Semax, Selank or placebo, and reported both shared and peptide-specific changes in functional connectivity between the right amygdala and the right temporal cortex11.
Human studies and regulatory status
Human research on both peptides is small and almost entirely Russian. For Semax, the clearest data are short imaging studies in healthy volunteers such as the head-to-head study above11. For Selank, two clinical studies of 62 and 60 patients with anxiety-related diagnoses compared it with the benzodiazepines medazepam and phenazepam, using standard rating scales and, in one, serum enkephalinase activity; neither used a placebo group12,13.
Russian authors describe Semax as a medicine used in Russia for patients with cerebral circulation disorders8. US pharmacists have described Selank as a poorly studied Russian drug with GABAergic mechanisms that was being sold to US consumers as a dietary supplement14. Belgian control-laboratory scientists who identified both peptides in seized preparations described them as research peptides sold online that, to their knowledge, had not completed clinical trials15. Neither peptide is approved by the FDA or the European Medicines Agency.
Limitations of the evidence
The two literatures share the same weaknesses. Most studies come from a small set of Moscow institutes, and the Semax and Selank papers often share authors. Endpoints are frequently mRNA levels measured hours after a single exposure in rodents, which show that a peptide changes gene expression but not whether those changes matter for function. Independent replication outside Russia is rare, the human studies are small, and several clinical papers are available only as Russian-language articles with short English abstracts.
- No specific receptor has been characterized for either peptide.
- Behavioral results come mostly from rodent stress and learning tasks.
- Clinical comparisons used active drugs rather than placebo.
- Regulators outside Russia have not evaluated either compound.
Choosing between Semax and Selank for research
The practical difference is the question being asked. Semax fits work on neurotrophin signaling, BDNF and ischemia models; Selank fits work on GABAergic modulation, enkephalin metabolism and stress behavior. Both are available individually as Semax and Selank, and together with Pinealon in the Semax / Selank / Pinealon Set.
Every Peptides Only lot is independently tested and ships from the USA with a lot-matched certificate of analysis showing HPLC purity and mass-spec identity, published in our certificate library. Our guide on how to read a peptide COA explains what those results mean.
Frequently asked questions
What is the difference between Semax and Selank?
Both are Russian heptapeptides ending in Pro-Gly-Pro. Semax is built on the ACTH(4-7) fragment and is studied mainly for BDNF expression and ischemia models1,7; Selank is built on tuftsin and is studied mainly for GABA receptor modulation, enkephalinase inhibition and stress behavior2,9.
Are Semax and Selank in the same peptide family?
In a sense. Both are glyproline hybrids that attach Pro-Gly-Pro to an active fragment3, but the fragments come from unrelated parent peptides: ACTH for Semax and tuftsin for Selank.
Are Semax and Selank approved by the FDA?
No. Neither is approved by the FDA or the European Medicines Agency. Semax is used clinically in Russia8, and Selank has been described as a Russian drug sold to US consumers as a supplement14. Peptides Only sells both strictly as research compounds.
Has anyone compared Semax and Selank directly?
Yes, in a few studies: an in vitro enzyme study in human serum10, a rat 6-OHDA model5 and a resting-state fMRI study in healthy volunteers11. Each found some shared and some distinct effects.
How should lyophilized Semax and Selank be stored?
Like most lyophilized peptides, sealed vials are generally kept cold, dry and protected from light. Our guide to peptide storage and stability covers the general principles.
