What “tested” means here
A single purity figure doesn't tell the whole story. Here is every check a lot passes before it ships, and what each one is for.
5 checks before a lot is released
A lot ships only after every check passes. The results go on its certificate.
Mass spectrometry confirms the molecule is what the label says.
HPLC measures the share of the target peptide in the lot.
LAL testing screens for bacterial endotoxin.
Culture testing checks for microbial growth.
ICP-MS screens for lead, arsenic, cadmium and mercury.
A purity number is only the start
HPLC tells you how much of the vial is the target peptide. It can't see these four things, so we test for them separately.
Endotoxin
Cell-wall debris left by bacteria during manufacture.
- What HPLC misses
- HPLC reads molecular purity. Endotoxin is biological, so a vial can pass HPLC and still carry it.
- Why it matters
- Even trace endotoxin provokes an inflammatory response in cell and tissue models, which skews results before an experiment starts.
Sterility
Whether anything is alive in the vial.
- What HPLC misses
- A clean chromatogram says nothing about bacteria, yeast or mould. That takes a culture test.
- Why it matters
- Microbial growth adds an uncontrolled variable to every assay the vial touches.
Heavy metals
Lead, arsenic, cadmium and mercury carried over from reagents.
- What HPLC misses
- Metals are inorganic and invisible to HPLC and mass spec. They need a separate ICP-MS run.
- Why it matters
- Metals interfere with enzymes and accumulate over long studies, confounding the data.
Lot consistency
Whether this lot behaves like the last one.
- What HPLC misses
- One vial per run is the usual spot-check. It cannot show drift between lots.
- Why it matters
- If the material drifts between orders, so do your results. Reproducibility depends on it.
